There are half dozen differences between single stage and multistage multiplexing but the most common one is that Single stage multiplexing means that HO ODU’s can be mapped only to the same type of OTUk, but multi-stage multiplexing allows that one HO ODUj is still mapped to another HO ODUk before mapping it to OTU.

Single stage multiplexing also helps inbandwidth fragmentation and improving the wavelength channel utilization whereas multistage doesn't do that.
