The main distinction between DNA polymerase 1 and 3 is that DNA polymerase one is involved with the removal of primers from the fragments and substituting the gap by relevant nucleotides. In contrast, DNA polymerase three is mostly involved in the synthesis of the leading and lagging strands.

DNA polymerase 1 is a polymerase encoded by the pol A gene and is implicated in the prokaryotic DNA replication. DNA polymerase 3 is the primary enzyme that assists in prokaryotic DNA replication. DNA belongs to the DNA polymerase one family A., while DNA polymerase 3 belongs to family C. DNA 1 acts on the lagging strand. DNA 3 acts on both leading and lagging strands of the replication.
