Progesterone levels peak a week after ovulation, that is day 21 in a 28 day menstrual cycle. It peaks several days after ovulation, and then falls back to low levels unless the woman gets pregnant. Serum progesterone is a test to measure the amount of progesterone in your blood. Serum progesterone level is usually checked six to eight days after you ovulate (about day 21 of a day 28 cycle).
