What is the equation for doing a cell count on fluids using a hematocytometer?
# of cells counted x (dilution x 10/# of squares) = cell count
How do you dilute a very bloody sample?
1:20, 950 microliters of diluent to 50 microliters of sample
If you see cells on the hematocytometer, what do you do next?
Do a differential
How is a differential performed on a fluid?
Cytospin 1-4 drops of fluid dispersing on amt of cells in the fluid and count 100 cells. Dilute the sample if necessary.
What are normal cells seen in a spinal fluid?
Lymphs, monos, choroid plexus cells, segs
What are cells that can be found in spinal fluid during a disease state?
Plasma cell, macrophage, lipophage, increased amount of segs, pyknotic neutrophils, eos, clusters = malignancy, bacteria, malignant cells
What are normal cells seen in pleural and pericardial fluid?
Mesothelial cells, segs, lymphs, monos, macrophage
What are cells that can be seen in pleural and pericardial fluid during a disease state?
Pyknotic cells, macrophage, LE cells, carcinoma cells, clumps = malignancy
What are normal cells seen in synovial fluid?
Synovial lining cells, segs, lymphs
What are cells that can be seen in synovial fluid during a disease state?
LE cells, crystals, clumps
What is the calculation for a manual WBC?
Number of cells counted x (20 x 10/4) = number of cells
How is a manual WBC done?
Dilute the blood by 20, 950 microliters diluent and 50 microliters of blood. count 4 corners, check with bottom and average
What is the calculation for nucleated rbcs?
A=BC/C+DA=WBC count corrected for nrbcsB=WBC countC=Constant (100)D=# of nrbcs
What tests are done to diagnose pernicious anemia?
Schilling's Test, B12/Folate studies, Intrinsic Factor antibody(also retic ct)
What tests are done to diagnose Hereditary spherocytosis?
Osmotic fragility (membrane protein studies may also be done?)
